As for your work colleague that's a touchy one. From personal experience it's not always good to mix business and pleasure.

Easiest thing to do is ask her out for a drink she'll else say yes if she's interested or no if shes not.

Not everyone is interested some girls just flirt, some guys read singles wrong.
